Unusual metallo-β-lactamases may constitute a new subgroup in this family of enzymes

چکیده
Metallo-β-lactamases (MBLs) are a family of Zndependent enzymes that have contributed strongly to the emergence and spread of antibiotic resistance. Novel members as well as variants of existing members of this family are discovered continuously, compounding their threat to global health care. MBLs are divided into three subgroups, i.e. B1, B2 and B3. The recent discovery of an unusual MBL from Serratia proteamaculans (SPR-1) suggests the presence of an additional subgroup, i.e. B4. A database search reveals that SPR-1 has only one homologue from Cronobacter sakazakii, CSA-1.These two MBLs have a unique active site and may employ a mechanism distinct from other MBLs, but reminiscent of some organophosphate-degrading hydrolases.

Evolution of Metallo-β-lactamases: Trends Revealed by Natural Diversity and in vitro Evolution
The production of β-lactamase enzymes is one of the most distributed resistance mechanisms towards β-lactam antibiotics. Metallo-β-lactamases constitute a worrisome group of these kinds of enzymes, since they present a broad spectrum profile, being able to hydrolyze not only penicillins, but also the latest generation of cephalosporins and carbapenems, which constitute at present the last resou...
متن کاملIdentification of 76 novel B1 metallo-β-lactamases through large-scale screening of genomic and metagenomic data
BACKGROUND Metallo-β-lactamases are bacterial enzymes that provide resistance to carbapenems, the most potent class of antibiotics. These enzymes are commonly encoded on mobile genetic elements, which, together with their broad substrate spectrum and lack of clinically useful inhibitors, make them a particularly problematic class of antibiotic resistance determinants. We hypothesized that there...
متن کاملAn Update on the Status of Potent Inhibitors of Metallo-β-Lactamases
The production of metallo-β-lactamases is the most important strategy by which pathogenic bacteria become resistant to currently known β-lactam antibiotics. The emergence of these enzymes is particularly concerning for the future treatment of bacterial infections. There are no clinically available drugs capable of inhibiting any of the metallo-β-lactamases, so there is an urgent need to find su...

Metallo-β-lactamases (MβL) catalyzing the hydrolytic cleavage of the four-membered β-lactam ring in broad spectrum of antibiotics and therefore inactivating the drug; However, the mechanism of these enzymes is still not well understood. Electronic structure and electronic energy of metallo-β-lactamase active center, two inhibitors of this enzyme including penicillin and cephalexin, and differen...
متن کاملEmergence of Imipenem Resistance and Presence of Metallo--Lactamases Enzymes in Multi Drug Resistant Gram Negative Bacilli Isolated from Clinical Samples in Kerman, 2007-2008
Background & Aims: Imipenem is a member of Carbapenem with stability against most -lactamases .It is of particular use in the treatment of infections associated with drug resistant gram negative bacteria harboring ESBL and AmpC genes. The aim of this study was to determine the imipenem resistance in gram negative bacteria causing nosocomial infections and the presence of metallo--lactamases (MB...
